Karki works for Mitini Nepal, an organisation supporting LGBTI rights. It was established in 2005 with the aim of protecting and promoting rights of sexual and gender minorities' people of Nepal. As a LGBTI activist, she regularly faces threats and intimidation from relatives of their client or village communities. She has been threatened to be raped, kidnapped or killed.
